The main function of cortisol is to increase blood glucose levels to stimulate the body. Its function, therefore, is primarily to give us more energy in situations where we are exposed to physical or emotional stress. The main function of ACTH is to regulate cortisol, a glucocorticoid (steroid) hormone as well as androgens released by the adrenal glands. ACTH (1-39) is a potent agonist of the endogenous melanocortin 2 (MC2) receptor (EC50 = 57 pM). A component of the hypothalamic-pituitary-adrenal (HPA) axis that stimulates the production and release of glucocorticoids from the adrenal cortex. Adrenocorticotropic hormone (ACTH), which belongs to the melanocortin group of chemical peptides, was first isolated in the 1940s by purifying it through salt fractionation from sheep samples. In mammals, the action of ACTH is limited to those areas of the adrenal cortex that produce the glucocorticoid hormones cortisol (hydrocortisone) and corticosterone. ACTH has little control over the secretion of aldosterone, the other major steroid hormone from the adrenal cortex.

Interestingly, several short peptide sequences of ACTH have been shown to accelerate the maturation of the central nervous system, improve neuronal regeneration, and facilitate memory consolidation and preservation. These findings suggest that ACTH may provide beneficial cognitive neuroprotective effects, in addition to its ability to control epileptic seizures and interictal abnormalities.
