Inflammation is not the cause, it't's the symptom. The kidney study that proves it.
Posted on: 06/12/2026Imagine a water filtration plant that processes 180 litres of blood per day, recovering vital nutrients while excreting waste with remarkable precision. That is the kidney. Like any complex industrial system, it degrades with age — and unlike the heart or liver, it has very limited regenerative capacity. By the time most people reach their 70s, kidney function has typically declined by 30–50% from its youthful peak, even in the absence of overt disease.
SLU-PP-332 and the Aging Kidney
A 2023 study published in the American Journal of Pathology (Wang et al., PMID: 37717940) delivered some of the most compelling data to date on SLU-PP-332's therapeutic potential — directly in aged kidneys.
The Problem: ERRs Decline with Age
The study's first striking finding was that ERRα and ERRγ expression is significantly decreased in the kidneys of both aging humans and aging mice. This decline correlates with increasing mitochondrial dysfunction, inflammatory cytokine accumulation, and oxidative stress — the biological triad that drives age-related kidney deterioration. Importantly, lifelong caloric restriction — the most consistently pro-longevity intervention across species — preserved ERR expression in aged mouse kidneys, suggesting that ERR signalling is a key mediator of metabolic resilience.
SLU-PP-332 Intervention: Eight Weeks, Significant Results
Twenty-one-month-old mice (equivalent to approximately 70-year-old humans) were treated with SLU-PP-332 for eight weeks. The results were notable:
Reduction in renal fibrosis — the progressive scarring of kidney tissue that marks irreversible functional loss.
Decreased expression of pro-inflammatory cytokines, including TNF-α and IL-6, suggesting a significant anti-inflammatory effect mediated through metabolic reprogramming rather than direct immunosuppression.
Restoration of mitochondrial function, including improved oxidative phosphorylation capacity in renal proximal tubular cells — the kidney's highest-energy-demanding cells.
Improved renal gene expression profiles, with upregulation of PGC-1α, TFAM, and fatty acid oxidation genes — a signature consistent with metabolic rejuvenation rather than symptomatic masking.
The Mechanism: Inflammation Driven by Energy Failure
This research illuminates a paradigm increasingly supported across organ systems: chronic inflammation in aging tissues is often a downstream consequence of energy failure, not its cause. When mitochondria malfunction, cells shift to anaerobic metabolism, produce excess reactive oxygen species (ROS), and trigger inflammatory cascades. By restoring mitochondrial function via ERR activation, SLU-PP-332 appears to break this cycle upstream — addressing the energy failure before it becomes inflammation.
In simple terms: if the kidney's power supply is restored, it stops sending distress signals that trigger inflammation.
Clinical Implications
Chronic kidney disease (CKD) affects approximately 850 million people worldwide and is a leading cause of cardiovascular mortality. Current treatments slow progression but cannot restore lost function. SLU-PP-332's ability to reduce fibrosis, inflammation, and mitochondrial dysfunction in aged kidneys — with eight weeks of treatment — represents a mechanistically novel therapeutic avenue. Future research must clarify the compound's safety profile in aging kidneys and determine optimal dosing windows.
